The principle of MALDI is to irradiate a co-crystal film formed by a sample and a matrix with a laser, and the matrix absorbs energy from the laser and transmits it to the biomolecules. Protons are transferred to or from biomolecules during ionization. The principle of TOF is that the ions are accelerated to fly through the flight tube under the action of an electric field, and the mass-to-charge ratio (M/Z) of the ions is determined according to the time to reach the detector.

RP-HPLC

RP-HPLC

RP-HPLC is a liquid chromatography system consisting of a non-polar stationary phase and a polar mobile phase. The typical stationary phase for RP-HPLC is octadecyl-bonded silica and the typical mobile phases are methanol and acetonitrile. RP-HPLC is a type of chemically bonded phase chromatography using a non-polar bonded stationary phase. In RP-HPLC, the polarity of the bonded stationary phase is less than that of the mobile phase to make it suitable for the separation of non-polar, polar or ionic compounds.

Use the difference in molecular weight of proteins in the sample to separate them in an electrophoresis gel. Since sodium dodecyl sulfate and polyacrylamide respectively eliminate the influence of protein structure and charge during electrophoresis, the migration rate of proteins is only determined by their size.
