Overview of Acidic Amino Acids-based VLP Modification

Amino acids are biologically important organic compounds consisting of amino (-NH2) and carboxyl (-COOH) functional groups, and side chains attached to each amino acid. Amino acids are the basic units that constitute proteins, which endow proteins with specific molecular structure and form, and enable their molecules to have biochemical activity. Acidic amino acids, which are amino acids containing two carboxyl groups and one amino group, are mainly glutamate and aspartate.

  • Aspartate- or Glutamate-based VLPs Modifications
    The chemical conjugation method for constructing cVLPs based on aspartate and glutamate modifications differs from the modification methods involving cysteine and lysine. First, aspartic and glutamic must be activated with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) because only after activation can their carboxyl groups react with n-hydroxysuccinimide (NHS) to form NHS esters. The carboxylic acid side chain can then form a stable amide bond with a ligand having a primary amino acid. This method is mainly used to embed peptides or fluorescent probes into VLPs to form cVLPs.
